摘要:

阿尔茨海默病(AD)是一种以进行性认知功能下降为主要特点的神经退行性疾病。铁死亡是一种与神经退行性疾病相关的新型细胞调控性死亡方式,其中包括AD。铁死亡的核心过程是脂质过氧化引起细胞死亡,而脂氧合酶(LOX)在介导脂质过氧化方面发挥了重要作用。此外,脂质机制/途径的失调与许多神经退行性疾病有关,AD就是其中之一。越来越多的研究表明,LOX与AD的关系密切,LOX表达水平在AD患者大脑中升高,对β-淀粉样蛋白(Aβ)及Tau蛋白代谢、突触完整性及认知功能有一定影响。本文旨在分析LOX在铁死亡驱动AD中的关键作用,重点包括LOX的代谢过程、LOX与AD的关系、LOX抑制剂对AD的作用。

Abstract:

Alzheimer’s disease(AD) is a neurodegenerative disease characterized by progressive cognitive decline. Ferroptosis is a novel cell-regulated mode of death associated with a variety of neurodegenerative diseases, including AD. Lipoxygenase(LOX) plays an important role in mediating lipid peroxidation, which is the core process of ferroptosis. In addition, impairment of lipid mechanisms/pathways is associated with many neurodegenerative diseases, of which AD is one. More and more studies have shown that LOX is closely related to AD, and the level of LOX increases in the brain of AD patients, which has a certain influence on the metabolism of amyloid beta(Aβ) and Tau proteins, synaptic integrity and cognitive function. This article aims to analyze the key role of LOX in AD driven by ferroptosis, focusing on the metabolic process of LOX, the relationship between LOX and AD, and the effect of LOX inhibitors on AD.
