S100A6 对人肺癌细胞株Calu-6 分化的影响研究

摘要:

目的 探讨S100A6对人肺癌细胞株Calu-6分化方向的影响。方法 本实验时间为2021年6月至2022年2月,将15只8~10周龄BALB/c裸鼠采用简单随机法分为S100A6实验组7只、空载体组4只、空白对照组4只。在前期实验基础上构建S100A6过表达慢病毒、空载体慢病毒,将S100A6过表达慢病毒、空载体慢病毒及空细胞分别注射到S100A6实验组、空载体组、空白对照组裸鼠右前背部。第2周开始每周测量裸鼠的肿瘤体积,5周后处死裸鼠,取出肿瘤组织。采用免疫组化印记法检测肿瘤组织中甲状腺转移因子(TTF)-1、天冬氨酸蛋白酶A(NapsinA)、P40、CK5/6、P63的表达情况。结果 第2、3、4、5周三组裸鼠肿瘤体积比较,差异有统计学意义(P <0.05);其中S100A6实验组裸鼠肿瘤体积小于空载体组和空白对照组(P <0.05)。三组肿瘤组织中TTF-1、NapsinA、P40、CK5/6均表达阳性,S100A6实验组、空载体组、空白对照组肿瘤组织中P63表达阳性率分别为7/7、3/4、3/4。结论 S100A6可抑制人肺癌细胞株Calu-6增殖,对人肺癌细胞株Calu-6向肺腺癌或肺鳞癌分化方向未见影响。

Abstract:

Objective To explore the effect of S100A6 on the differentiation of human lung cancer cell line Calu-6. Methods This experiment lasted from June 2021 to February 2022. Fifteen BALB/c nude mice aged 8-10 weeks wererandomly divided into three groups, 7 in S100A6 experimental group, 4 in empty vector group and 4 in blank control group.S100A6 overexpression lentivirus and empty body lentivirus had been constructed base on previous experiment, and S100A6overexpression lentivirus, empty body lentivirus and empty cells had been injected into the right anterior back of nude micein S100A6 experimental group, empty body group and blank control group respectively. The tumor volume of nude mice wasmeasured weekly from the second week. The nude mice were killed after 5 weeks to remove the tumor. The expression ofthyroid transfer factor (TTF) -1, aspartate proteinase A (NapsinA) , P40, CK5/6 and P63 in tumor tissues was detected byimmunohistochemistry. Results There was a significant difference in tumor volume among 3 groups at week 2, week 3, week 4and week 5 (P < 0.05) ; the tumor volume of nude mice in S100A6 experimental group was smaller than that of empty body groupand blank control group (P < 0.05) . TTF-1, NapsinA, P40 and CK5/6 were positive in tumor tissues of the three groups, and thepositive rates of P63 in S100A6 experimental group, empty body group and blank control group were 7/7, 3/4 and 3/4 respectively.Conclusion S100A6 can inhibit the proliferation of human lung cancer cell line Calu-6, but has no effect on the differentiationof human lung cancer cell line Calu-6 into lung adenocarcinoma or lung squamous cell carcinoma.
