急性缺血性脑卒中患者血清 Janus 蛋白酪氨酸 激酶 2 和细胞因子信号转导抑制因子 1 水平 及其与肠道菌群、免疫功能的关系研究

摘要:

 目的 分析急性缺血性脑卒中(AIS)患者血清Janus蛋白酪氨酸激酶(JAK)2、细胞因子信号转导 抑制因子1(SOCS1)水平及其与肠道菌群、免疫功能的关系。方法 选择2020年2月至2023年2月宜昌市中心人民医 院收治的AIS患者106例为AIS组,另选取同期在宜昌市中心人民医院进行体检的健康者106例为对照组。比较两组血 清JAK2 、SOCS1水平和肠道菌群(乳酸杆菌、双歧杆菌、大肠埃希菌、肠球菌)数量及免疫功能指标( CD3 + 、CD4 + 、 CD8 + T淋巴细胞百分比);采用 Pearson相关分析探讨AIS患者血清JAK2、SOCS1水平间的相关性及二者与肠道菌群数 量、免疫功能指标的相关性。结果 AIS组血清JAK2水平高于对照组,SOCS1水平低于对照组(P< 0.05 )。 AIS组乳 酸杆菌、双歧杆菌数量低于对照组,大肠埃希菌、肠球菌数量高于对照组(P<0.05)。AIS组CD3 + 、 CD 4 + T淋巴细胞 百分比低于对照组,CD8 + T淋巴细胞百分比高于对照组(P<0.05)。Pearson相关分析结果显示,AIS患者血清 JAK2 水平与血清SOCS1 水平呈负相关( P<0.05);AIS患者血清JAK2水平与乳酸杆菌数量、双歧杆菌数量、 CD3 + T淋巴细 胞百分比、CD4 + T 淋巴细胞百分比均呈负相关,与大肠埃希菌数量、肠球菌数量、 CD8 + T淋巴细胞百分比均呈正相关 (P<0.05);AIS患者血清SOCS1水平与乳酸杆菌数量、双歧杆菌数量、 CD3 + T淋巴细胞百分比、 CD4 + T淋巴细胞百分 比均呈正相关,与大肠埃希菌数量、肠球菌数量、CD8 + T淋巴细胞百分比均呈负相关( P<0.05)。结论 AIS患者血 清JAK2水平升高,SOCS1水平降低,且二者间呈负相关;此外,血清JAK2、SOCS1水平均与肠道菌群数量及免疫功能 指标存在相关性,即随着血清JAK2水平升高、SOCS1水平降低,乳酸杆菌数量、双歧杆菌数量、 CD3 + T淋巴细胞百分 比、CD4 + T淋巴细胞百分比逐渐降低,大肠埃希菌数量、肠球菌数量、CD8 + T淋巴细胞百分比逐渐升高。

Abstract:

Objective To analyze the serum levels of Janus kinase (JAK) 2, suppressor of cytokine singaling 1 (SOCS1) and their relationship with intestinal flora and immune function in patients with acute ischemic stroke (AIS) . Methods A total of 106 AIS patients admitted to Yichang Central People's Hospital from February 2020 to February 2023 were selected as the AIS group, and 106 healthy subjects who underwent physical examination in Yichang Central People's Hospital during the same period were selected as the control group. Serum JAK2 and SOCS1 levels , intestinal flora (lactobacillus, bifidobacterium, Escherichia coli, enterococcus) and immune function indexes (CD3 + , CD4 + , CD8 + T lymphocyte percentage) were compared between the two groups. Pearson correlation analysis was used to investigate the correlation between serum JAK2 and SOCS1 levels and their correlation with intestinal flora and immune function indexes in AIS patients. Results The serum JAK2 level in AIS group was higher than that in control group, and the serum SOCS1 level was lower than that in control group (P < 0.05) . The number of Lactobacillus and bifidobacterium in AIS group was lower than that in control gro up, and the number of Escherichia coli and enterococcus was hig her than that in control group (P < 0.05) . The percentage of CD3 + and CD group, and the percentage of CD8 + T lymphocytes was higher than that in control group (P < 0.05) . Pearson correlation analysis showed that serum JAK2 level was negatively correlated with serum SOCS1 level in AIS patients ( P < 0.05) ; serum JAK2 level in AIS patients was negatively correlated with Lactobacillus number, bifidobacterium number, CD3 + T lymphocyte percentage, CD 4 + T lymphocyte percentage, and positively correlated with Escherichia coli number, enterococcus number, and CD8 + T lymphocyte percentage (P < 0.05) ; serum SOCS1 level in AIS patients was positively correlated with Lactobacillus number, bifidobacterium number, CD3 + T lymphocyte percentage, CD 4 + T lymphocyte percentage, and negatively correlated with Escherichia coli number, enterococcus number, and CD8 + T lymphocyte percentage ( P < 0.05) . Conclusion In AIS patients, the serum JAK2 level is increased and SOCS1 level is decreased, and there is a negative correlation between them. In addition, serum JAK2 and SOCS1 levels are correlated with the number of intestinal flora and immune function indicators, that is, with the increase of serum JAK2 level and the decrease of SOCS1 level, lactobacillus number, bifidobacterium number, CD3 + T lymphocyte percentage and CD 4 + T lymphocyte percentage gradually decrease, the Escherichia coli number, enterococcus number, and CD8 + T lymphocyte percentage gradually increase. 4 + T lymphocytes in AIS group was lower than that in control
